چکیده

While investigating the effects of structural analogues of thymine on the thymine-requiring strain of E&cherichia coli 15 T we noticed the presence in the bacterial deoxyribonucleic acid of a new base which we have identified as 6-methylaminopurine. Although 6-methylaminopurine is usually present in small amounts (2% of the adenine) in the deoxyribonucleic acid of E. coli 15 Tits relative proportion increases considerably during growth of the bacteria in thymine-deficient conditions, obtained either by maintaining the thymine concentration in the medium at a low level, or by the addition of thymine antagonists such as 5-aminouracil or 2-thiothymine. Examination of a number of deoxyribonucleic acids has shown that 6-methylaminopurine also occurs in small proportions as a constituent of deoxyribonucleic acid from several bacteria and bacterial viruses. This paper describes the identification of 6-methylaminopurine, its deoxyribonucleoside and deoxyribonucleotide from deoxyribonucleic acid of E. coli 15 T, and the occurrence of the base in other deoxyribonucleic acids. Brief accounts of this work have appeared previously (Dunn & Smith, 1955a, b).

and in mice bearing Ehrlich ascites tumor has been investigated. The results indicate that the exogenous compound is utilized equally well for the synthesis of both adenine and guanine of deoxyribonucleic acid and ribonucleic acid. No evidence for the incor poration of intact 6-methylaminopurine into the nucleic acids of liver, kidney, spleen, intestine, or tumor cells was obtained. The princip...

Occurrence of Methylated Purine Bases in Yeast Ribonucleic Acid*

A number of methylated purine bases have recently been reported to occur in small quantities in the urine of normal human subjects. These include 1-methylguanine, iV*-methylguanine (6-hydroxy-2-methylaminopurine) ,I 1-methylhypoxanthine, and 8-hydroxy-7-methylguanine (1) in addition to 7-methylguanine, previously known as a urinary constituent (2). These compounds were found to be metabolites o...

The occurrence and distribution of thymine and three methylated-adenine bases in ribonucleic acids from several sources.

It has been known for several years that deoxyribonucleic acid may normally contain 5-methylcytosine (Wyatt, 1951), 5-hydroxymethylcytosine (Wyatt & Cohen, 1953), and 6-methylaminopurine (Dunn & Smith, 1955, 1958a) in addition to adenine, guanine, cytosine and thymine. The first evidence of such additional bases naturally present in ribonucleic acid is the recent isolation of an unidentified nu...

The occurrence of methylated guanines in ribonucleic acids from several sources.

A number of additional purines and pyrimidines have recently been described as minor constituents of ribonucleic acids. Cohn (1957, 1958, 1959) and Davis & Allen (1957) have described an unusual pyrimidine nucleotide from yeast ribonucleic acid, thymine, 2-methyladenine, 6-methylaninopurine and 6-dimethylaminopurine have been identified as components ofribonucleic acid from several sources (Lit...
